Mostly Expression of Interest (EOI) is valid for at least one year from the date applicants submit it to the MPNP. If after completing one year in Manitoba you did not receive a Letter of Advice to Apply (LAA) but you still want to live and work in the province, then you will have to create or complete a new profile. Making changes to the old profile will not help you to get LAA.
You cannot apply to the MPNP if you are one of the following:
1. Spouse or common-law partner of Canadian Permanent Residents or Canadian Citizens
2. A federal appeal or removal process involving refugee claimants.
3. Temporary overseas workers who are currently residing and working in a province of Canada other than Manitoba.
4. Applicants whose applications have been refused by MPNP in the past 6 months.
5. Applicants with active immigration applications with the federal government or any other province.

If you are a Provincial Nominee, you will surely get issued a WPSL (Work Permit Support Letter) by the MPNP, you just have to make a written request for WPSL in person or via mail. After receiving a Work Permit Support Letter (WPSL) if you want you can apply to IRCC (Immigration Refugees and Citizenship Canada) for a Work Permit.
Yes, you can apply for the Manitoba PNP without the Federal Express Entry for all programs, besides Express Entry program under the Manitoba Skilled Worker Overseas program or stream.
